ABSTRACT
HLA‐B*15:753 differs from HLA‐B*15:579 by one nucleotide substitution in codon 73 in exon 2.

We report here a novel HLA‐B*15 allele, now named B*15:753 that carries one nucleotide substitution in exon 2 when compared to the B*15:579 allele, identified in a volunteer haematopoietic stem cell donor. The HLA typing was performed using Next Generation Sequencing reagents (NGSgo‐MX6‐1, GenDX, Utrecht, Netherlands) run on the Miseq system platform (Illumina, San Diego, CA). The reads were analysed using the NGSengine software (version 4.0) The subject was found to have a new B*15 allele and was consequently typed as HLA‐A*02:01, 68:01; ‐C*07:02, 12:02; ‐B*15:753, 52:01; ‐DRB1*11:01, 14:04; ‐DQB1*03:01, 05:03; ‐DPB1*02:01, 04:01. Using the IPD‐IMGT/HLA Database [1], nucleotide sequence alignment with HLA‐B alleles shows that this new allele has one nucleotide change from B*15:579 in codon 73 of exon 2, where A→G, resulting in a coding change (codon 73: ACT→GCT, Threonine→Alanine, Figure 1). This nucleotide change was confirmed using Next Generation Sequencing reagents provided by One Lambda (AllType NGS, Canoga Park, CA) run on the Miseq system platform (Illumina, San Diego, CA). The reads were analysed using the TypeStream Visual Software version 3.1 (One lambda, Canoga Park, CA). The nucleotide sequence of the new allele has been submitted to the GenBank database (accession number PX395728) and to the IPD‐IMGT/HLA Database (Submission No. HWS10101493). The name B*15:753 has been officially assigned by the WHO Nomenclature Committee for Factors of the HLA System in October 2025. This follows the agreed policy that, subject to the conditions stated in the most recent Nomenclature Report [2], names will be assigned to new sequences as they are identified. Lists of such new names will be published in the following WHO Nomenclature Report.
