Effect of dexamethasone on IL-5 promoter constructs. (a) Primary human T cells, transiently transfected with the constructs indicated, were incubated with PMA (50 nm) + PHA (5 μg/ml). Cells were harvested at 12 hr and luciferase activity was measured. Data are expressed as means ± SEM of at least four independent experiments. (b) Primary human T cells were transiently transfected with either of the two IL-5 promoter constructs and stimulated as in (a) and co-incubated with dexamethasone (1 μm) and harvested as above. Data are expressed as fold induction as means ± SEM of at least four independent experiments. (c) Supernatants from the above experiments were harvested and IL-5 release was measured.
