AcGFP-tagging and presence of rhesus macaque β2-microglobulin do not affect the surface expression of rhesus macaque MHC class I molecules. a Constructs coding for Mamu-A1*00101, A1*00801, A1*01101, A2*050402, A3*1311, A4*1403, B*01202, B*02101, B*02804, B*06002, I*010201 and I*0121 either with or without C-terminal AcGFP were transiently transfected in K562 cells. Surface expression was measured 2 days post-transfection by flow cytometry with W6/32 and polyclonal PeCy5-coupled goat anti-mouse antibody. The MFI ratios of AcGFP-tagged versus untagged MHC class I of four independent experiments are indicated. Horizontal bars indicate the average and vertical bars denote SEM. b K562 cells were transiently transfected with AcGFP-tagged Mamu-B constructs along with either none (control), 2 µg, or 3 µg of rhesus macaque β2-microglobulin expression construct (mmB2M). Expression of mmB2M was controlled in parallel experiments with anti-V5 tag-specific antibody. The average of three independent experiments is shown and error bars denote SEM
