Depletion of most exosome subunits does not affect cell proliferation. (A) S2 cells were treated with exosome subunit specific dsRNAs as described in Materials and Methods. Effectiveness of dsRNA treatment was determined by Western blotting and depletion was calculated by comparing bands with a loading standard (Lamin Dm0). Representative blots for cell proliferation experiments are shown for each depleted exosome subunit. (B) During dsRNA treatment, cells were counted by hemacytometer at designated times. (C) S2 cells were depleted and counted as above. On day five, a portion of the cells were transferred to fresh flasks and treated with dsRNA for an additional 5 d. The graphs represent the average of three independent depletions for each subunit tested.
