Thermal stabilization of a solubilized GPCR (hKOR1) by introducing cholesterol analogs into DDM detergent micelles: DDM (●), DDM/CHS (▲), DDM/CHOBIMALT (○). The mass ratio of 1% DDM to the cholesterol analog was 5:1. Thermal stability was assessed at 30 °C in 50 mM Tris pH 7.4, followed by radioligand binding at 5 °C. Specific binding of the radiolabeled antagonist 3H-diprenorphine was measured as a function of incubation time in order to determine the half-life for retention of receptor function upon gradual thermal inactivation. Half-lives determined for hKOR1 in are: DDM 141 ± 5 minutes, DDM/CHS 247 ± 10 minutes, DDM/CHOBIMALT 267 ± 8 minutes.
