Abstract
We isolated Mesoflavibacter zeaxanthinifaciens S86 as xylanase-producing bacteria from seawater sampled in Micronesia. Analysis of the M. zeaxanthinifaciens genome revealed that it contains a single circular chromosome of 3,704,661 bp with 3,249 putative open reading frames.
GENOME ANNOUNCEMENT
Experimental bacterial samples were collected from seawater of Chuuk State in Micronesia. The collected samples were cultured in an XYS agar plate (0.2% xylan, 0.2% yeast extract, and 1.5% agar in natural seawater) at 30°C. Xylan-degrading bacteria was selected for identification to the species level. 16S rRNA gene sequence analysis revealed that the strain is 99.4% similar to the 16S rRNA gene sequence of Mesoflavibacter zeaxanthinifaciens (accession no. AB265181); hence, it was named M. zeaxanthinifaciens S86. M. zeaxanthinifaciens is a Gram-negative, rod-shaped, mesophilic bacterium that belongs to the family Flavobacteriaceae of the phylum Bacteroidetes (1). A zeaxanthin-like carotenoid with industrial potential has been isolated from M. zeaxanthinifaciens, indicating that this bacteria has remarkable potential for various commercial applications, such as pigmentation of foodstuffs, cosmetics, and pharmaceuticals (1).
The M. zeaxanthinifaciens S86 genome was sequenced by Roche GSFLX 454 technology (Macrogen), which revealed a total of 122,643,197 bases and a total of 304,049 reads with an average read length of 808 bp. The genome contained 296,787 assembled reads and 3,611 partial assembled reads. The reads were assembled into 57 large contigs (length, >500 bp) and 76 small contigs (length, >100 bp) using genome sequencer FLX system De Novo software v. 2.3. The average contig size was 64,974 bp, and the genome contained 3,249 predicted coding sequences. Results showed that the genome consists of 499 singletons. The complete genome of M. zeaxanthinifaciens S86 is on a single circular chromosome of 3,704,661 bp with an average G+C content of 37.1%. Additionally, the M. zeaxanthinifaciens S86 genome contains 22 rRNA operons.
Genes coding for saccharification enzymes were detected in the M. zeaxanthinifaciens S86 genome. There were 39 different saccharification enzymes corresponding to four types of xylanases. Xylanases can catalyze the hydrolysis of β-1,4-xylosidic linkages in xylan to produce xylooligosaccharides and xylose (3). Xylanases have a broad range of potential applications in the textile, waste treatment, animal feed, paper, and biofuel industries, suggesting that the M. zeaxanthinifaciens S86 genome contains valuable data for commercial applications (2).
