Heparan sulphate (HS) -stimulated production of cytokines in RAW264.7 cells was inhibited by paxilline. (a) RAW264.7 cells were incubated for 4 hr at 37° with HS (1 μg/ml) or lipopolysaccharide (LPS; 10 ng/ml) in the absence and presence of paxilline (2 μg/ml), respectively. After centrifugation, the concentrations of tumour necrosis factor-α (TNF-α) in the supernatants were analysed using an ELISA kit. (b) RAW264.7 cells received the same treatment as mentioned above. After 12 hr, the concentrations of interferon-β (IFN-β) were determined by ELISA.
