E2-stimulated protein SNO was enhanced by overexpression of mitochondrial or Golgi, but not membrane- targeted eNOS in HUVECs. Mitochondrial eNOS (Mito-eNOS), CAAX-eNOS, and wt-eNOS were overexpressed in HUVECs, respectively. After treatment with or without 10 nM E2β, cells were lysed and subjected to biotin-switch. The biotin-labeled SNO-proteins were analyzed by immunoblotting with antibiotin antibody. Expression levels of eNOS were determined with anti-eNOS antibody.
