Selective deletion of COX-2 in myeloid-COX-2 KO mice. BMDM were stimulated with lipopolysaccharide (5 µg/ml, 6h) to induce COX-2 expression. COX-2 was significantly reduced in myeloid-COX-2 KO compared with WT BMDM by (A) Q-PCR for mRNA (Left, n = 6) and quantification of COX-2 peptide by liquid chromatography/mass spectrometry in MRM mode (Right, n = 5–7), whereas (B) COX-1 expression was unaltered. (C) Lipopolysaccharide-induced PGE2 synthesis in myeloid-COX-2 BMDM was abolished and PGD2 markedly decreased compared with WT (n = 3–4). (D) Constitutive (no lipopolysaccharide) COX-2 mRNA levels were not altered in kidney and brain tissue of myeloid-COX-2 KO compared with WT mice (n = 4). RQ = relative quantity.
