Shad1 modification and correlation of expression with YB-1. (A) The Shad1 RNAs possess a 5′-phosphate as indicated by a 5′-phosphate-dependent exonuclease reaction. (B) Periodate oxidation followed by polyadenylation and qPCR reveals 3′ modifications in YB-1–associated RNAs and the positive control, miR-135m, a synthetic oligo with a 3′ end 2′-O-methyl modification. (C) Genomic location of Shad1 shyRNA and smyRNA as determined by sequencing of YB-1–associated RNAs. (D) In a normal prostate epithelial cell line (RWPE), YB-1 expression is less than that of the prostate cancer cell line (PC-3). Shad1 level is also down-regulated in RWPE compared with PC-3.
