Whole cell lysates of PC-3 and DU 145 cells treated with the indicated concentrations of 3-AWA and vehicle were subjected to western blot analysis to determine (A) phosphorylation of MTOR and AKT along with total MTOR, total AKT, and loading control ACTB. (B) ER stress sensor proteins HSP90B1, HSPA5 and the proapoptotic protein PAWR. (C) PC-3 cells were treated with the indicated concentrations of 3-AWA in the presence or absence of the ER stress inhibitor salubrinal (50 μM) to determine the expression of EIF2AK3 and LC3B-I/-II by western blot analysis.
