Immunodetection of core fucosylated glycoproteins by Western blot with PhoSL lectin following PSA immunoprecipitation (IP). A) Total PSA IP with Hybritech antibody, B) Free PSA IP with M30 antibody from Roche and C) double IP procedure firstly with Hybritech antibody followed by IP with M30 antibody from Roche. IP starting material consists of 500 µl of pooled female sera spiked with 100 ng (panels A and B) and 200 ng (panel C) of standard (Std) PSA. As a reference, 50 ng and/or 100 ng of Std PSA were loaded in each gel, as well as the molecular markers (identified in panel A). The arrow at the right side indicates the band corresponding to PSA.
