(A) PRL-3 overexpression increased phosphorylation of STAT-3 (Y705) and total levels of STAT-1 in human myeloma cell line INA-6-WT. Cells washed 4 times and depleted of IL-6 for 3 hours before incubation without or with 10 pg/mL or 1 ng/mL IL-6 for 30 minutes. (B) INA-6-WT cells were starved for 3 hours, pre-incubated with inhibitor for 20 minutes before addition of IL-6. DMSO was used as control in inhibition experiments to exclude DMSO effects.
