In the article listed above the line separating panels A and B of Figure 2 was placed incorrectly, through an image in panel A. A corrected Figure 2 appears below. The publisher apologizes for this error.
Figure 2.
Assembling two kb-long RNA molecules by ligation using RNA substrates generated by site-specific RNase H cutting. (A) 2′-O-methyl RNA (4 nt) - DNA (4 nt) - 2′-O-methyl RNA (4 nt) oligonucleotide annealing guides RNase H to cut an RNA molecule site-specifically.19 Three internal positions (‘•’, ‘◊’ and ‘♦’) on the 1 kb renilla luciferase mRNA or the 1.9 kb firefly luciferase mRNA were chosen for RNase H cutting. (B) RNase H cutting for all three positions is 93–99% complete (lanes 7–9). The residual uncut full-length mRNA is accounted for in the ligation efficiency calculation (Materials and Methods). Addition of DNA proximal disruptors to the ligation reaction increases the yield of full-length mRNA from 2% (lane 1) to 14% (lane 2) for the 1 kb renilla luciferase mRNA, and from 0% (lanes 3 and 5) to 9–10% (lanes 4 and 6) for the 1.9 kb firefly luciferase mRNA. The gel is denaturing acrylamide-urea gel.
