Genetic removal of histone modifier genes affects W/O switching. All homozygous deletion mutants were constructed in the MTL heterozygous recipient wild-type strain J4-2.1 (WT MTLa/α). Strains were grown on YPD plates at 30°C for 2 days. Then W cells were plated on Lee’s GlcNAc medium at 25°C with 5% CO₂ for 5 days to quantify W/O switching. Frequencies were calculated as follows: [(opaque + opaque-sectored colonies)/total colonies] × 100. *, P < 0.05, and **, P < 0.01, compared to the WT strain.
