Complement-mediated HCV binding to Raji cells. (A) HCV genotype 1a virus in 3 ml medium was incubated with 100 µl serum sample or medium only at 25°C for 1 h, followed by mixing with 2 ml of Raji cells (1×10⁶ cells/ml) and incubating at the indicated temperature for 1 h. (B) 2 ml of Raji cells (1×10⁶ cells/ml) were incubated with 10 µg each of the indicated antibody at room temperature for 30 min, followed by mixing with 3 ml of complement-activated HCV genotype 1a virus (pre-incubation with serum for 1 h at 25°C, 1×10⁷ genomic copies per reaction), and incubating at 25°C for 1 h. After incubation, the cells were pelleted, washed two times with complete RPMI medium, and processed for RNA isolation and HCV quantification as described in Methods section. Each value represents the mean ± SD of 6 determinations.
