Gating strategy to define lymphocyte subsets. (A) Representative dot plots of the gating strategy. Lymphocytes were first gated according to the forward and side scatter profile. Doublets were excluded from the analyses using side scatter area vs width discrimination. Dead cells, which were determined by positive staining for the cell viability dye, were then excluded. (B) CD3 then CD4 and CD8 markers were used to determine the lymphocyte primary populations, which were then gated into subsets according to the corresponding fluorescence-minus-one (FMO) control (middle and bottom panels).
