(A) Baseline collagen genes (COL1A1, COL1A2 and COL3A1) were observed. TU-BcX-2O0 have higher levels of COL1A1 and COL1A2 compared to COL3A1. (B) MDA- MB-231 cells treated with LBH589 (100nM, 24h) show reduction of COL1A2 and increase of COL3A1 mRNA expression. (C) TU-BcX- 2O0 tumors treated with LBH589 from the in vivo treatment experiment have a significant reduction of COL1A2, similarly observed with MDA-MB-231 cells. COL3A1 mRNA expression were undetectable in all samples, and could not be analyzed. All data was obtained using qRT-PCR and normalized to actin. Error bars represent SEM and significantly different * p < 0.05, *** p < 0.001. Data from tumors was performed in quadruplicate; all other data was performed in triplicate.
