P1 (the residue of the substrate) specificity refers to primary substrate specificity, which is determined by primary substrate-binding pocket called S1 binding sites of the protease. Extended substrate specificity is determined by secondary substrate-binding pockets, called S2 and S3 binding sites of the protease.
