Tyrosine-phosphorylated cortactin (yellow P) does not associate with srGAP1. Steady-state signaling at junctions may activate a kinase that maintains most cortactin in a phosphorylated state. HGF leads to dephosphorylation of cortactin, allowing recruitment of srGAP1. srGAP1 in turn decreases Rho activity via its GAP domain, thereby destabilizing cadherin-dependent cell–cell adhesion.
