IgG-opsonization of Friend murine leukemia virus (F-MuLV) diminishes activation of virus-specific CD8 T cells by bone marrow-derived dendritic cells (bmDCs) but not by B cells. 5 × 105 (A) DCs or (B) B cells were loaded with 5000 FFUs of F-MuLV or an equivalent of C’- or IgG-opsonized F-MuLV (F-MuLV-C or F-MuLV-IgG, respectively) based on viral RNA. DCs or B cells were cocultured with 5 × 105 isolated FV-specific TCRtg CD8 T cells. Activation of CD8 T cells was determined by the expression of the early activation marker CD69 gating on AAD-negative living CD8 singlets measured by flow cytometry after 48 h of coculture. Bars represent mean ± SEM of five independent experiments. Data were analyzed by GraphPad PRISM (version 7) software (GraphPad Software, San Diego, CA, USA) using ANOVA followed by Dunnet’s multiple comparison test (***, **, * significant at p < 0.001, p < 0.01, p < 0.05, respectively).
