The study design and single-strand library preparation of cfDNA in conception. cfDNA fragments were first denatured into single-strand DNA (ssDNA) fragments. Then, the ssDNA fragments were ligated with a unique molecular identifier. The pre-library was enriched using hybridization and magnetic-beads capture and then subjected to high-throughput sequencing.
