Effect of PMA and bryostatin 1 on Munc13-1 and bMunc13-2 membrane translocation. The top panel shows representative Western blot analysis of the cytosolic (C) and membrane (M) fractions of Munc13-1 and bMunc13-2 after cells were treated with PMA or bryostatin 1 (1 μM) for 5 min. The bottom panel bar graph shows normalized densitometric analysis of immunoblots from multiple experiments, of which the top panel is representative. Data are expressed as means ± SEM of four independent experiments. One-way ANOVA, followed by Tukey’s post hoc test, was used for analysis of statistical significance. *P < 0.05 compared to the vehicle control for Munc13-1 and bMunc13-2. **P < 0.01 compared to the vehicle control for Munc13-1 and bMunc13-2. HT22 cells were transiently transfected with Munc13-1 and bMunc13-2, and Western blot analysis was carried out as described in Materials and Methods.
