Physiological parameters for the P. pastoris strains. Physiological parameters of three P. pastoris strains expressing CH3H as well as one empty strain. Standard deviations were calculated from triple measurements and calculated by error propagation. Differences in the maximum specific methanol uptake rates were determined significant between Mut+ and MutS strains at a confidence level of 95 %. In the SMD1168H cultivation strong methanol accumulation occurred after 48 h of induction. Therefore, the feed‐rate was reduced. However, the biomass concentration continuously decreased as a result of feeding close to the maintenance level. This resulted in an extremely low biomass yield and high standard deviation thereof.
