Effects of gintonin on insulin secretion from INS-1 cells and rat pancreatic islets. (a,b) Cells were preincubated in Krebs-Ringer bicarbonate (KRB) buffer (0.2 mM glucose) for 1 h before incubation with gintonin. (a) Cells were incubated with the indicated concentrations of gintonin (GT) in KRB buffer (3.3 mM glucose or 16.7 mM glucose) for 2 h. (b) Cells were incubated with gintonin (30 μg/mL) in KRB buffer (3.3 mM glucose or 16.7 mM glucose) for the indicated periods. (c) Rat pancreatic islets were preincubated with RPMI1640/F12 containing 5.6 mM glucose for 1 h and subsequently incubated with the indicated concentrations of gintonin in RPMI1640/F12 containing 5.6 mM glucose for another 1 h.
