Isoharringtonine (IHT) inhibits the growth of non-small cell lung cancer (NSCLC) cells in 2D and 3D culture systems. (A) A549 and NCI-H460 cells were seeded in ULA plates and grown for 2 days before IHT treatment with the indicated concentrations. After 72 h, cell viability was measured by the CellTiter-Glo 3D cell viability assay kit. *, significantly different from control (p < 0.05). (B) 2D-cultured cells were exposed to IHT at the indicated concentrations for 48 h, and cell viability was examined by the CellTiter-Glo assay. *, significantly different from control (p < 0.05). (C,D) The median effective dose (ED50) values of IHT were obtained from dose-response curves generated by GraphPad Prism. (E,F) Representative images were collected before viability assay. Scale bars, 100 μm. All data are presented as mean ± SD.
