Characterization of the phenotype of antigen-specific T cells in the lung parenchyma of immunised mice and controls at 8 and 20 weeks post vaccination. Mice were vaccinated as shown in Fig. 2a. Lung cells were assessed 8 weeks post vaccination using flow cytometry and presented as cell number (1, 2). (a) Ag85A-specific lung parenchymal CXCR3+ KLRG1- CD4+ T cells and (b) Ag85A-specific lung parenchymal CD44hi CD69+ CD11a+ CD4+ T cells at 8 weeks after vaccination. (c) Ag85A-specific lung parenchymal CD8+ T cells expressed CXCR3+ KLRG1-. (d) Ag85A-specific lung parenchymal CD8+ T cells expressed CD44hi CD69+ CD103+. A duplicate experiment was analysed at 20 week-time point. Lung parenchymal CD4+ and CD8+ T cells were phenotypically classified using the memory makers and presented in the same order (e-h). Each symbol represents one animal and the line is the median of each group (n = 3–6 mice). Data are representative of three independent experiments.
