TPP inhibits the synthesis of viral nsp2 in infected Marc-145 cells. (A) Marc-145 cells were infected with PRRSV-CHR6 (MOI = 0.6) for 6 h, then different concentrations of TPP were added, and cells were incubated for another 36 h. IFA for the nsp2 protein of PRRSV was performed using Alexa-Fluor-488-conjugated anti-mouse secondary antibody (green). The actin protein was stained with Alexa-Fluor-555-conjugated anti-rabbit secondary antibody (red). Nuclei were stained with DAPI (blue). Scale bar, 100 μm. (B) The mCherry-tagged nsp2 plasmid was transfected into Marc-145 cells for 6 h, and cells were then mock-treated or treated with TPP for 24 h. Cells were finally harvested for Western blot analysis.
