Figure 1 Schematic representation of the different methods used to modulate sEVs with a Cy3‐labelled miRNA mimic and the follow‐up assays performed to validate the modulation and assess the bioactivity of the modulated sEVs. Five different methodologies have been used to load miRNAs into sEVs: transfection by Exo‐Fect or cholesterol‐modified miRNA and membrane permeabilization by a detergent (saponin), electroporation or heat shock (1). The modified sEVs were then characterized regarding their loading efficiency by fluorescence and qRT‐PCR analyses (2), bioactivity in the HEK‐293T reporter cell line (3), cell toxicity using a cell viability assay (4) and capacity to transfect cells (5).
