MIO genes are required for capsule growth under inositol-inducible conditions. (A) Wild type H99 and its mio1Δ2Δ3Δ triple mutant were incubated on capsule-inducing minimal medium with glucose (MMD) or inositol (MMI) as the carbon source for 3 days before photography. (B and C) Capsule size and cell body size of H99 and its mio1Δ2Δ3Δ mutants cultured in glucose (Glu), inositol (Ino), or both (Glu/Ino) were analyzed by measuring over 100 cells. Statistical analysis was done using two-tailed Student's t test. ****, P < 0.0001. (D) Detection of capsule using FITC-labeled GXM monoclonal antibody 18B7. Wild-type H99 and its mio1Δ2Δ3Δ mutants were cultured on MM for 3 days before coincubation with the GXM monoclonal antibody 18B7. The acapsular cap59Δ mutant was used as a control.
