The 3 enzyme assays used to detect the 2 enzyme activities of NDST enzymes. 1” N-deacetylase activity is measured in an ELISA assay where the monoclonal antibody JM403 recognizes the free amino-groups generated after N-deacetylation. 2” N-sulfotransferase activity is measured after incorporation of ³⁵S-sulfate groups from [³⁵S]PAPS into the deacetylated K5-polysaccharide substrate. 3” combined N-deacetylase and N-sulfotransferase activities are measured after incorporation of ³⁵S-sulfate groups from [³⁵S]PAPS into the K5-substrate.
