Cytogenetic and phenotypic comparisons of proliferation rate and clonogenicity potential of NHSM derived and isogenic primed hESCs at early and late passages

(A) Proliferation rate was determined by the doubling time assay.

(B) Fraction of cells in the S-phase was analyzed using EdU incorporation and quantitative DNA staining followed by FACS analysis.

(C) Clonogenicity potential determined by alkaline phosphatase staining of colonies 7 days after seeding at low density. All data are expressed as mean ± SEM, ∗p < 0.05, ∗∗p < 0.01, ∗∗∗p < 0.005 by a paired t-test.

(D) Karyotype analysis of derivation (p20) hESC lines.
