The May 10, 2021, article by Gillessen et al entitled “Predicting Outcomes in Men With Metastatic Nonseminomatous Germ Cell Tumors (NSGCT): Results From the IGCCCG Update Consortium” (J Clin Oncol 10.1200/JCO.20.03296) was published with errors.
While working on a new manuscript based on their data, the authors were informed by one of the contributors to the IGCCCG-Update project that there were errors in the assignment of the values of the variable “site of primary” for 113 of their patients. 48 patients initially reported as having gonadal primary had in fact retroperitoneal (12) or mediastinal (36) primary; 12 patients initially reported as having a retroperitoneal primary had in fact gonadal (10) or mediastinal (2) primary; 34 patients initially reported as having mediastinal primary had in fact gonadal primary (32) or mediastinal primary (2), and one patient is now excluded from the analysis set as having missing primary site.
Importantly the data from that contributor was used in the model development set. The correction leads to slight changes to the estimates of the model parameters since 94 of the 3,542 patients included in the prediction model development set are affected by the change. Consequently, the prediction model slightly changed: The conclusions of the paper have not changed.
The Hazard Ratio (HR) of mediastinal primary versus other increased from 2.68 to 3.44.
The HR of presence versus absence of NPVM (with AFP ≤ 30 ng/mL, HCG ≤ 5 U/L, LDH/UNL ≤ 2.5) decreased from 6.61 to 5.98.
The HR of presence vs absence of lung metastasis increased from 1.62 to 1.66.
The HR of LDH > 2.5 × UNL vs ≤ 2.5 × UNL in the presence of NPVM decreased from 1.01 to 0.88
The HR of doubling of AFP levels for value > 30 ng/mL in the presence of NPVM decreased from 1.02 to 1.00 (from very little impact on risk of progression or death to no impact on risk of progression or death according to the model)
The other HRs were not affected
As a result of the change to the model, all the figures that document model parameters, all figures and tables in the Data Supplement required changes except for Supplementary Table 2. The Data Supplement has been replaced and relabeled as Data Supplement 1.
Table 1, Table 3, Figure 1, Figure 2, and Figure 3 have been replaced.
Data Supplement 2 has now been provided to show details of reproducing the IGCCCG-Update model prediction.
The web app (https://www.eortc.org/IGCCCG-Update) has been corrected accordingly.
