The two bands in the T. tauschii HindIII digest were likely due to an internal HindIII in the gene. The band sizes (HindIII, 7.1 kb; EcoRI, 4.4 kb; BamHI, 6.9 kb) in rice agreed with expected sizes from genome sequence (GenBank accession no: AP000364).
