Fig. 6. BMS-806 exerts minimal effects on sCD4 binding to cell-surface HIV-1 envelope glycoproteins. FITC-soluble CD4 (sCD4) (10-30 m g/ml) was incubated with 293T cells expressing the cytoplasmic tail-deleted YU2 HIV-1 envelope glycoproteins, in the presence of the indicated concentrations of BMS-806. After washing, the cells were analyzed by fluorescence-activated cell sorting. Background binding of FITC-sCD4 to mock-transfected cells was determined in parallel experiments and subtracted from the values obtained for envelope glycoprotein-expressing cells. The results are expressed as the percentage of FITC-positive cells observed in the presence of BMS-806 relative to that observed in its absence. The results represent the means and SDs obtained in four independent experiments.
