  • Movie 1 - Video S1. This video shows a live-cell imaging time course of internalized Cy3-STxB in wild-type HAP1 cells. The time is shown in minutes in the top left hand corner. Each time point is a maximum intensity projection of the whole cell taken as a series of Z slices. Notice how after only 10-15 minutes there is observable accumulation in the juxtanuclear region of the cells, representing the Golgi complex.
  • Movie 3 - Video S3. This video shows a live-cell imaging time course of internalized Cy3-STxB in TSSC1-KO HAP1 cells. The time is shown in minutes in the top left hand corner. Each time point is a maximum intensity projection of the whole cell taken as a series of Z slices. Notice how after an hour Cy3-STxB is found in vesicles dispersed within the cytoplasm, comparable to the VPS54-KO cells.
  • Movie 2 - Video S2. This video shows a live-cell imaging time course of internalized Cy3-STxB in VPS54-KO HAP1 cells. The time is shown in minutes in the top left hand corner. Each time point is a maximum intensity projection of the whole cell taken as a series of Z slices. Notice how after an hour Cy3-STxB is found in vesicles dispersed within the cytoplasm.
